Leave the M25 near the Dartford Crossing and follow the A13 east, then the signs for the Port of Tilbury. Your cruise documents say which gate and terminal to use.
Parking
Cruise parking is booked ahead, at or near the terminal. Book it when you book the cruise, and take the details from your cruise line.
By air
London City and London Southend are the nearest airports. Heathrow, Gatwick and Stansted are all reachable by road around the M25.
